Twin Wolves, Twin Hearts is a subquest in the The Oni's quest. It starts after the "A Fiery Consequence" quest, where Atsu and Jubei in the end decide to fight together against the Oni.
Twin Wolves, Twin Hearts: Step-by-step guide
Step 1 - Meet Jubei in the Matsumae camp
Travel to the northern Matsumae camp to meet Jubei, who is with the commander Wada. Enter the tent to trigger a cutscene.
Step 2 - Travel with Jubei
Follow Jubei and travel to the Azure Wind Shrine to find Mad Goro. You will have to find a way to reach the shrine as there are obstacles encompassing the path.
Step 3 - Find a way to the shrine
After reaching the shrine, you observe the bounty hunters around the area. Go to the left side and climb the cliff with Jubei. Continue the path until you come across a bunch of Ronin in the area.
Step 4 - Kill the group of ronins
Kill the Ronins as it would help you to find location ahead without any hassle of being targeted from behind. You can kill them via silent assassination or direct fight.
Step 5 - Find the path ahead
The bridge has been burnt by fire and can't be used. Head to the left and try to jump across with the help of a rope. You can later jump towards the cliff near the bridge and climb up.
Step 6 - Remove the debris on the bridge
Clear the debris using your rope to make it accessible for Jubei to cross and then continue your way to the shrine.
Step 7 - Travel through the cave
The path you follow leads you inside the cave. Destroy the wooden fence and follow Jubei, you will end up falling into a different path once you cross the bridge.
Step 8 - Climb across the mountain
After exiting the cave, climb or grapple your way up to meet with Jubei.
Step 9 - Reach the summit
Continue climbing to the top of the summit where shrine is located. You will see Mad Goro welcoming you.
Step 10 - Honor the shrine
Mad Goro asks you to follow the shrine rules by washing your hands and mouth and paying respect to the shrine. This will give you the Charm of Hachiman.
Step 11 - Talk to Mad Goro
Mad Goro gives you the oni flames as the waves of bounty hunters appear on the scene.
Step 12 - Defeat the Ronins
Defeat the ronins as they surround the shrine. Use Oni's Flame to kill the enemies for additional assitance. Else, fight as you like.
Step 13 - Continue further down the cliff
As you make your way down the shrine while defeating the enemies further more will join, around three waves enemies join the fight. Continue downwards until you see the horses.
Step 14 - Mad Goro reveals his secret route to The Oni's castle
As you end the fight and a cutscene triggers where Mad Goro tells Atsu and Jubei the secret location and how to get there. This is what you wanted from him. The subquest ends here.

This sub quest is pinitol to the bonding of the Atsu and Jubei and finding a path towards defeating the Oni. It requires stealth and swiftness of climbing.
